Adding a new layer to cancer immunotherapy

TME-anchored CD28 costimulation designed to enable a truly effective immune response against cancer. Concept covered by multiple patents, ready for partnering.

Our Market

Solid tumors remain a challenge for T cell-based immunotherapy.

T cell engagers deliver powerful immune activation against cancer, but treatment of solid tumors comes with challenges including insufficient T cell activation and an immunosuppressive TME.

What if the tumor microenvironment could become an enabler rather than a barrier to a powerful and sustained immune response?

TWYCE is developing a strategy that combines tumor targeting with TME-anchored costimulation to drive a potent and more sustained T cell response in solid tumors.

Overcoming the solid tumor challenge.

Solid tumors remain a challenge for T cell-based immunotherapy. Amongst others, an immunosuppressive tumor microenvironment can limit T cell access and activity, while the lack of truly tumor-specific targets may lead to toxicity and restrict dosing. New approaches need to combine precise targeting with potent, durable immune activation.

The next step beyond CD3 T cell engagement.

CD3 T cell engagers have established tumor-directed T cell activation as a powerful therapeutic approach, with major success in hematologic malignancies and growing validation in solid tumors. BiCos that provide costimulation specifically within the TME will critically enhance both the durability and the specificity of the induced T cell response.

Targeted costimulation built for combination

We are developing BiCos as TME-anchored CD28 costimulatory antibody constructs designed to pair with internal CD3-based programs as well as cancer vaccines or partner-provided tumor-targeting approaches. By localizing costimulation to the TME, BiCos drive potent immune activation while limiting systemic CD28 activity. Accordingly, BiCos may serve as a combinatorial partner for immunotherapy in a broad spectrum of malignant disease.

Platform & Value Creation

One platform, multiple combinations across targets, tumor types and modalities.

TWYCE combines TME-anchored CD28 costimulation with tumor-targeting programs and rapid translational validation. Ongoing CC-1 (PSMA×CD3) and CC-3 (CD276×CD3) clinical studies provide information on safety and dosing as well as biological insights for future combinations with BiCos, while vaccines and next-generation formats broaden the opportunity.
